Pfizer Reports Positive AA Trial Results – A JAK3 inhibitor and a tyrosine kinase inhibitor (TYK2/JAK1) have shown statistically significant results in a phase 2a trial conducted by Pfizer. The company announced the results on September 15, 2018 at the European Academy of Dermatology and Venerology Congress. Subjects of the trial received oral doses of the drugs over a 6 month period. The TYK2/JAK1 inhibitor showed the greater efficacy, improving hair regrowth by 49.5 points on the Severity of Alopecia Tool scale, compared to an improvement of 33.6 points by the JAK3 inhibitor. However, Pfizer has apparently decided to move forward with its JAK3 inhibitor due to 2 adverse events in the TYK2/JAK1 inhibitor cohort during the trial. Pfizer’s JAK3 candidate, PF-06651600, was also recently granted Breakthrough Designation from the US FDA for treating alopecia areata.

Researchers from the NIH and the University of Alabama, Birmingham have discovered a connection between the body’s innate immune regulation and hair graying. It was found that the transcritpion factor known as MITF, which plays an important role in melanocyte function, also plays a major role in hair graying. When the body’s immune system is dealing with a pathogenic infection such as bacteria or virus, molecules called interferons will send out signals to the body to take action against the pathogen. If MITF loses control of interferon response in melanocyte stem cells (due to an immune system response), hair turns gray. Essentially, immune system response may contribute to the process of hair graying according to the study.

It’s not clear at this time whether WAY-316606 is approved and on the market, or if it was partially developed to treat the bone disease.
The Holy Grail remains a drug that will promote regrowth, but this might not be so far away. Earlier this year, Manchester University announced that an osteoporosis drug had been found to have “dramatic results” promoting hair growth when applied to tissue samples in pre-clinical trials. The resultant frenzy left the PhD student responsible, Dr Nathan Hawkshaw, a little dazed. “Every other week, something comes out about hair loss and it doesn’t generate as much media coverage as what I experienced,” he grumbles. McElwee is an associate professor in the Department of Dermatology and Skin Health at the University of British Columbia (UBC) in British Columbia, Canada and director of the Hair Research Laboratory in the Vancouver Coastal Health Research Institute (VCHI) at Vancouver General Hospital (VGH). A hair research scientist, McElwee is one of only a small group of research scientists worldwide who studies hair biology and associated diseases.
Dr. Hawkshaw and his team were lead to test WAY-316606 for hair growth after studying the effects of cyclosporine A (CsA) on hair growth. They found that CsA reduced the expression of SFRP1 in human hair follicles. After looking for other drug candidates that had a similar effect on SFRP1, WAY-316606 was identified. The team has already tested WAY-316606 on isolated human hair follicles which were donated from hair transplant surgeries, and plans to test the drug in human clinical trials in the future. A timeline for a human clinical trial has not been set yet, Follicle Thought will update this as news is presented.

Hair transplantation involves harvesting follicles from the back of the head that are DHT resistant and transplanting them to bald areas. A surgeon will remove minuscule plugs of skin that contain a few hairs and implant the plugs where the follicles are inactive. Around 15 percent of hairs emerge from the follicle as a single hair, and 15 percent grow in groups of four or five hairs.


All that said, our primary consideration for choosing hair loss shampoos for our list was whether it produced actual results. We also acknowledge that what might work for someone may not work for another, but we’re interested in the overall picture. Perhaps not surprisingly, the “big names” in the hair loss game – Lipogaine, Nizoral, etc. – made the grade, in part, because they’ve accumulated years of successful results.

Hair grooming, but more importantly, having a head-full of hair is as important to men as it is to women. To women, it may be an important accessory of beauty, and for men, it adds to a sense of manliness, enhances their looks and makes them more appealing and attractive to women. Balding to men is associated with aging (only old men are expected to lose hair) and therefore, having hair on one's head is a sign of virility and masculinity.
With those pinned down, it wasn’t hard to determine which don’t actually work. Pretty much all the “active” ingredients listed in ineffective treatments — from biotin and zinc to emu oil and saw palmetto — have never been proven, and are instead marketed based on logical-seeming correlations. It would make sense that biotin, a B vitamin readily found in hair, skin, and nails, could help hair grow more quickly. And caffeine is a stimulant that works in coffee, so rubbing some on your scalp might wake some of those sleepy follicles… right?
1. Collagen powder. Preliminary studies suggest that marine-sourced collagen may stimulate hair growth, says New York City dermatologist Yoon-Soo Cindy Bae. While they prepared them for implantation, Bernstein explained the real future of the business: cloning. Bernstein has partnered with a Columbia University geneticist, Angela Christiano, who is working on duplicating hairs. The problem with hair transplantation is that you’re moving hairs around, not creating new ones. Minoxidil (Rogaine). Minoxidil is one of the most common and popular forms of medication for hair loss, particularly pattern baldness, but also generally used in cases of alopecia areata. It is an over-the-counter drug that comes in either liquid or foam form and is applied on the scalp, particularly on the bald patches, to promote hair growth and stop further hair loss.
Disruptions in the normal length of each phase, which can cause hair loss and hair thinning, may be the result of a number of internal and external stimuli. These are also what we call the triggers and causes of your hair loss. As a quick example, dieting can leave the body stressed and in need of important nutrients. Because of this stress, hair growth may be cut shorter than usual and there is an early onset of telogen or shedding of hair.

Hair loss is often distressing and can have a significant effect on the patient's quality of life. Patients may present to their family physician first with diffuse or patchy hair loss. Scarring alopecia is best evaluated by a dermatologist. Nonscarring alopecias can be readily diagnosed and treated in the family physician's office. Androgenetic alopecia can be diagnosed clinically and treated with minoxidil. Alopecia areata is diagnosed by typical patches of hair loss and is self-limited. Tinea capitis causes patches of alopecia that may be erythematous and scaly and must be treated systemically. Telogen effluvium is a nonscarring, noninflammatory alopecia of relatively sudden onset caused by physiologic or emotional stress. Once the precipitating cause is removed, the hair typically will regrow. Trichotillomania is an impulse-control disorder; treatment is aimed at controlling the underlying psychiatric condition. Trichorrhexis nodosa occurs when hairs break secondary to trauma and is often a result of hair styling or overuse of hair products. Anagen effluvium is the abnormal diffuse loss of hair during the growth phase caused by an event that impairs the mitotic activity of the hair follicle, most commonly chemotherapy. Physician support is especially important for patients in this situation.
As with the Nioxin shampoo, the PhytoCayne shampoo is meant to be used in conjunction with another product, the company’s revitalizing treatment serum. The shampoo is designed to “prepare” the scalp to absorb the serum. For that reason, you’ll find that this shampoo doesn’t really moisturize your scalp in the way the first two entries on our hair loss shampoo list do. What it does, though, is thicken hair for a fuller appearance, and many users say that it also significantly slows additional hair loss and breakage.
David made this claim back in 2000. But fast-forward a few years and his enhanced compensation strategy begins to look a little quaint. Androgenetic alopecia, or male pattern baldness, afflicts about half of all men aged 50 and they can’t all reinvent the sitcom. This particular research which was published in the Nature journal showed that a synthetic version of sandalwood, called Sandalore, binds to the OR2AT4 receptor in hair folliles and prolongs their anagen (growth) phase. The hair follicles studied were treated in a petri dish. Paus has subsequently announced that a completed clinical trial of 20 female volunteers using a topical version of Sandalore showed a reduction of daily hair loss. There is also another larger clinical trial ongoing now which hopes to confirm the effect and announce results in early 2019. Paus has gone as far to say “there is even a chance that this might be able to bring the hair back.” We’ll keep our fingers crossed.
Finasteride has limitations though, such as the requirement of daily treatment, a limit to how many damaged hair follicles it can revive, and that it may lose its effectiveness overtime for some people. This drug has shown to be better at preventing further hair loss than reversing it (regrowth). Just keep in mind that some side effects might make the hair loss seem more appealing.
